Description:
Phytoextraction of heavy metals is a promising technology to remediate slightly and moderately contaminated soils. To enhance crops' uptake of heavy metals, chelates such as EDGA are being tested as soil additives. Heavy metal loaded EDGA can affect soil organisms such as bacteria and nematodes in various ways: directly via the soil solution surrounding the organisms and indirectly by changing the approachability, amount and quality of specific food items for nematodes and bacteria. In a pot experiment with various crops growing in slightly polluted acid sandy soil (pH 4.5, 2mg Cd and 200mgZnkg^-^1 soil), Cd and Zn loaded EDGA in the soil solution did not affect herbivorous nematodes but did strongly reduce the increase in bacterivorous nematodes. Moreover, while the crop-stimulated increase in numbers of bacterivorous nematodes dropped, the measured amounts and the growth of their food (bacteria) were not reduced. This differential effect of the EDGA addition occurred under moderate (grass) and strong (lupine and yellow mustard) stimulation of bacterivorous nematodes by the crops, and of moderate (grass, yellow mustard) and no (lupine) stimulation of herbivorous nematodes. We assume that EDGA addition did not increase the load of bacteria with adsorbed heavy metals. Probably the bacterivorous nematodes were inhibited to feed by the high concentration of heavy metal-complexed EDGA in the soil solution, also surrounding their prey (bacteria). Although EDGA addition to the soil stimulated uptake of heavy metals by the crops, it decreased heavy metal concentrations in the roots. Herbivorous nematodes were therefore not negatively affected by the EDGA addition to the soil. Fungivorous nematodes were negatively affected by EDGA addition, probably due to increased heavy metal concentrations in the fungal hyphae. Thus, EDGA can have significant side effects on the functioning of the soil organisms, and a thorough analysis of trophic relationships among soil organisms is needed to understand these effects. ... The best microbiology book I've ever read (I haven't finished yet but it's still fun). I think it is the most up to date textbook in microbiology. It covers both biochemical and signal transduction pathways/networks inprokaryotes as well as principles in systematics and short introductions tomedical microbiology and biotechnology. At the end of every chapter thevarious authors provide references to essential papers and reviews (insteadto other books, like in Brocks Biology of Microorganisms) to start gettingfamiliar with journals. The graphics are done with less colours but arevery professional and as "easy" to understand as the topic theyare representing (chemotaxis is not as easy as described in the stryer orbrock). Maybe it's too much to read for undergraduates, but very useful forgraduate students. ... Read more

Description:
Microscopic studies have shown that most microbes exist in soils as dwarf cells that are thought to be an adaptation to nutrient limitation. Most dwarfs are uncultured by current approaches and have not been identified phylogenetically. Only a few dwarf bacteria have been isolated in culture and dwarf archaea have received little study. We selected a semiarid creosotebush shrubland site for studying dwarf microorganisms because arid and semiarid soils are generally nutrient-poor. Soils were collected beneath creosotebush canopies and in open areas between shrubs. Cells were eluted in sodium pyrophosphate and filtered with a 0.45-@mm pore-size filter. Filtrate DNA was extracted, PCR-amplified using universal bacterial and archaeal 16S rDNA primers, cloned, RFLP-screened, and sequenced. The eluted cell filtrates were also inoculated into R2B medium. After incubation, cultures were filtered to select against populations of dwarfs that formed large cells in the presence of nutrients (pleomorphic dwarfs) and to select for populations that retained dwarf size (intrinsic dwarfs). Dwarf archaea and bacteria were present in the initial filtrate and in the cultures. A single dwarf archaeon (SevArch-01) that is related to other soil Crenarchaeota sequences was found in the initial filtrates and in subsequent filtrate cultures, indicating an intrinsically dwarf archaeon. Dwarf bacteria fell into four bacterial phyla: Proteobacteria, Firmicutes, Actinobacteria, and the TM-7 group. Intrinsically dwarf bacteria in enrichment cultures were identified as @a- and @b-Proteobacteria. Dwarf bacteria related to Arthrobacter, Propionibacterium, and other actinobacteria were detected. This is the one and only text book I have ever read from cover-to-cover because it was interesting enough to hold my attention, and because it was written in such a way that the sections which discussed areas of bacteriology already known to me became useful exercises in revision.As a bacteriologist entering the complex world of eukaryotic cell signalling for the first time, this has been an invaluable text for me.The authors are to be congratulated on writing a text book on a subject which at times can be highly complex - since they have made the subject matter accessible to all, from grad students to research professors.The diagrams are without exception clear and concise, and the chosen literature for further reading is well-sourced. My only criticism is that some of the diagrams may have been easier to decipher if presented in colour - but I suspect the publishers were trying to keep printing costs to a minimum.

I do hope the authors consider producing updated revisions of this text as this field of microbiology progresses. ... Read more
