Extractions: The Genome carries all the genetic information found in every cell of an organism. It is often called "the blueprint for life". It is extremely tiny, and can only be seen with a microscope. It consists of all the deoxyribonucleic acid (DNA) of any organism. DNA is composed of two strands wrapped around each other, assembled like a twisted ladder (which is called a double-helix). Each strand is composed of repeating units called nucleotides. There are four different nucleotides in DNA: adenine (A), cytosine (C), guanine (G), and thymine (T). The two strands of DNA are held together by links between parallel bases, creating base pairs (bp), ( Casey, Denise ). Adenine pairs only with thymine and cytosine with guanine. The order of the nucleotides (bases) along the DNA strand is called a DNA sequence. An organism's DNA sequence carries the genetic instructions required to form another organism with the exact same traits. A gene is a specific sequence of bases that can vary in length. The sequence contains the instructions for building proteins. Each set of three base pairs codes for one amino acid, and each gene codes for a protein (proteins are made up of amino acids). For instance, if an average-sized gene contains 3,000 base pairs, the protein will include 1,000 amino acids. A molecule called messenger ribonucleic acid (mRNA) transmits the protein-coding instructions. The Human Genome is estimated to include more than 30,000 genes, according to the recent studies. Only ten percent of DNA comprises protein-coding sequences of genes. The rest is called "junk DNA".(Laura Mazzola, PhD.)

Deletions or duplications in the gene can lead to muscular dystrophy. Analysis of specific deletions provides information about the gene structure in individual cases. PCR of exons 21 and 34 of the dystrophin gene using QIAGEN Taq DNA Polymerase and Q-Solution gave highly specific results, allowing analysis of the gene structure in two muscular dystrophy patients. Another Taq DNA polymerase gave a large number of nonspecific bands (see figure). DNA from two muscular dystrophy patients (B, C) oand one unaffected individual ( A ) was analyzed by PCR using QIAGEN Taq DNA Polymerase and Q-Solution or Taq DNA polymerase from Supplier X following suppliers' specifications. PCR was carried out for 35 cycles using primers specific for a 178 bp fragment from exon 21 or a 102 bp fragment from exon 34 of the dystrophin gene. Patient C shows a deletion in both exon 21 and exon 34; patient B shows a deletion only in exon 21.

Extractions: Genetic Testing for Huntington Disease Facts About Finding the HD Gene, DNA Banking and Genetic Testing for Huntington Disease In 1983, a major breakthrough was made in the search for the cause of Huntingtons Disease. A DNA marker for the HD gene was found by Dr. James Gusella, of Massachusetts General Hospital. Although this marker was not the gene itself, its discovery meant that scientists were close to locating the gene. Subsequently, many other markers were found. Finding these markers made it possible to offer a presymptomatic test to persons who were at-risk for HD. However, not everyone could use this test because it required that an at-risk person have blood samples available from extended family members, both affected and unaffected. Our recommendation at that time was that regardless of whether or not a family member had decided to use testing, families should go ahead and store samples from family members who were affected, elderly or crucial to the testing process. After nearly ten years of effort, an international team of scientists headed by Dr. Gusella has identified the gene which causes HD. It took an intense scientific effort and the phenomenal cooperation of families to accomplish this task. Along with finding the gene, comes a host of new questions:
