DNA palindromes are a source of instability in eukaryotic genomes but remain under-investigated because they are difficult to study. Nonetheless, progress in the last year or so has begun to form a coherent picture of how DNA palindromes cause damage in eukaryotes and how this damage is opposed by cellular mechanisms. In yeast, the features of double strand DNA interruptions that appear at palindromic sites in vivo suggest that a resolvase-type activity creates the fractures by attacking a palindrome after it extrudes into a cruciform structure. Induction of DNA breaks in this fashion could be deterred through a Center-Break palindrome revision process as investigated in detail in mice. The MRX/MRN likely plays a pivotal role in prevention of palindrome-induced genome damage in eukaryotes. ... Read more

Recently, it has emerged that palindrome-mediated genomic instability contributes to a diverse group of genomic rearrangements including translocations, deletions, and amplifications. One of the best studied examples is the recurrent t(11;22) constitutional translocation in humans that has been well documented to be mediated by palindromic AT-rich repeats (PATRRs) on chromosomes 11q23 and 22q11. De novo examples of the translocation are detected at a high frequency in sperm samples from normal healthy males, but not in lymphoblasts or fibroblasts. Cloned breakpoint sequences preferentially form a cruciform configuration in vitro. Analysis of the junction fragments implicates frequent double-strand-breaks (DSBs) at the center of both palindromic regions, followed by repair through the non-homologous end joining (NHEJ) pathway. We propose that the PATRR adopts a cruciform structure in male meiotic cells, creating genomic instability that leads to the recurrent translocation. ... Read more

The poem that takes up the larger part of this book is a poem based on the fact that Columbus was looking for India when he stumbled upon the Americas.It is a fascinating exploration of this serendipitous discovery.But for me, the most incredible part of this book is the second piece."Palindrome Apocalypse" is a masterpiece.The Croatian text, which is placed opposite of the English translation is fascinating just to look at.Dubravka Oraic-Tolic has told a very difficult and emotional story through the use of palindromes, words/phrases that are the same forwards and backwards.I couldn't help just staring at the genius of the palindromes.The translation is also masterfully done with some very evocative thoughts about the suffering that Croatia has endured over the years.This is a book that will be a joy to look back on repeatedly.It is a great way to learn more about a country that is becoming more and more popular to visit.

I edited this book and designed the interior (with help). Usually I wouldn't comment on the book in a review because obviously I'm biased in favor of the book. I'm going to break that rule here though. I was not a fan of poetry. I prefer simple reading. But once I got into Dubravka's poetry, I could not help but feel amazement at the eloquence and quality of her poetry. The multiple layers of depth that intertwine subjects, times, and languages as if they were one, in fact contending in some ways they are. The language can be serious and humorous almost simultaneously. The insights are wonderful. Each time I read the poems (and I have read them hundreds of times) I still feel delight and newness. Dubravka is certainly one of the most intelligent people I have ever associated with. Her skill with written language is in the realm of genius. In story chronology, this is the first of a trilogy featuring villain Roberto Trebor, who loves to be unlovable and in fact lethal.He toys with his victims and taunts his pursuers, cleverly dangling wordplay clues that both lead and elude.
But it's not the bad guy with the outsized brain we love to follow, so much as the husband & wife team who alternately falter and figure out the zig-zag pathway Roberto's criminal mind has them traversing.We fall in love with the love between these professional investigators, cheer "Rosie" & Edna Rosenberg on and, ultimately, share their triumph.

Jeb Ladouceur has produced another thriller featuring the brilliant maniac, Roberto Trebor, a palindrome-fixated serial killer with no heart whatsoever. Matter of fact, Trebor finds it easy to cut out his victims' hearts (or throats) with aplomb.
"The Palindrome Plot" is actually the first installment in a trilogy involving homicide detective Steve "Rosie" Rosenberg and his battle with the seemingly indestructible fiend who torments him and his savvy sidekick, Edna Murphy. Trebor is fresh from jail, having been released by an errant parole board even though he has murdered his parents just a few years earlier. The initial offering in the three-book series was the memorable "Frisco" and bound-to-be-controversial "Calamity Hook" (rumored to flirt with sacrilege) is on the way soon.
Ladouceur is an interesting writer who really does his research homework. For instance, the inventive author makes his antagonist, Roberto, a custodial employee of the New York Public Library, and places him in the famed building's cavernous sub-basement. His makeshift room can be reached only by riding an ancient freight elevator that no other employee would ever use. There Trebor lures a youthful victim and ... well, I mustn't reveal the lurid details, but you may rest assured the goings on will punctuate your calm with absolute dread.
Unquestionably, Jeb Ladouceur has a fine future presenting his multi-layered plots and breathtaking tie-ins, for he does so with superb imagery and sure-footed linguistic style. So get "The Palindrome Plot" ... I dare you. For a while, late at night, you might be sorry you did ... but you'll probably get over it.
