Preparation of the following publications drew on the resources and/or facilities of the Intermountain Herbarium to some extent. The list is incomplete, as I have not been tracking such publications sufficiently diligently of late. The listing starts with 1996. Web Pages are listed separately because they are probably updated at irregular intervals. Some may have become extinct. Mary Barkworth, Director.

Eswaran, A. Saini, and N. Jawali (Molecular Biology and Agriculture Division). For combining desirable HMW-glutenin subunits and durable rust-resistance genes, intervarietal crosses involving recent bread wheat cultivars and some of our experimental lines were made. The cultivars and experimental lines used in the crosses were PBW-343, HUW-206, WH-542, HD-2385, HD-2285, Vidisha, KS-1, and SK-1. The F2 populations were scored for morphology, flowering time, and rust reaction after artificial inoculation. A study of genetic diversity of Indian cultivars using PCR-based markers is being investigated. The association of molecular markers with quality traits, agronomically important traits, and rust resistance genes also are being studied.

Australia Boosts Funding for Weed Management
Clearly, Australia is serious about weed management and is putting up the funding to prove it. The country's government recently announced A$77 million funding for four Cooperative Research Centers (CRCs), including the new CRC for Australian Weed Management (CRCAWM), previously known as the CRC for Weed Management Systems. The monetary support is to commence in July this year and continue through 2008. The government's announcement of the awards noted that "Weeds are amongst the most serious threats facing Australia's primary production and biodiversity with costs estimated at more than A$1.2 billion per year for grain cropping and probably at least twice that for the rest of agriculture." The CRCAWM will be working to reduce the risks to the environment, agriculture, and rural sectors all across Australia from currently established weed species as well as an increasing number of newly invading weeds.

Bacterial typning and subtyping using PCR/DGGE
Division of Microbiological Safety, Institute of Food Safety and Toxicology,
Multiple methods are applied to bacterial genotyping , but only one application have been published, where mutation detection by denaturing gradient gel electrophoresis (DGGE) were used on Chlamydia trachomatis omp1 gene have been published (Sayada et al., J.Med.Microbiol. 43, 14-25, 1995),
We have applied PCR/DGGE to Campylobacter jejuni subtyping using a flaA PCR fragment (702bp) as polymorphic target. Thirteen different subtypes were identified in an unrelated collection of 80 Campylobacter jejuni isolates. The disciminitory Index (0.896) was slightly better than serotyping (0.868), but significant lower than PFGE and RAPD (0.974 and 0.984, respective). Improvement might be obtained by heteroduplex formation. The PCR/DGGE method does not need any restriction enzyme digestion, and the band pattern is very simple to evaluates, as the position of only a single band in the gel indicate the genosubtype. Furthermore the modified version TTGE can be automated and performed in capillarY electrophoresis.
Simulation software WinMelt (BioRad) or Melt94, MUTRAV, and SQHTX (Lerman and Silverstein) are important tools to mimic the Tm melt-curves and mobility curves in denaturing gels of known DNA sequences. Our presentation will show to use of these simulations to optimize condition for DGGE mutation analysis. However, only empiric trials can be done on unknown samples.

BioCentrum, Technical University of Denmark, Bygn. 223, Lyngby 2800 Denmark Chromosome walk between genes methG, uvsB, hisA, gdhB and frA of A. nidulans produced two continuous segments of a physical map of chromosome IV covering 250kb and 160 kb. Integration of selectable markers into the chromosome targeted by unique cosmid fragments made it possible to establish that the two fragments flank the centromere, with the order of markers methG-uvsB-CEN-hisA-gdhB-frA . The gap in the physical map over the centromere itself was estimated to be between 70 kb and 250 kb. It was shown that the frequency of meiotic recombination in the vicinity of the centromere is strongly reduced. Pericentric fragnets contain highly repetitive AT-rich DNA and degenerated retrotransposons. Centromere-proximal DNA fragments were subcloned in circular and linear vectors, and their behavior in transformation was studied. n ) has also accelerated. In tandem mass spectrometry experiments, selected ions are decomposed by collisions with gases, photons, electrons or surfaces, and the resulting product ions yield information on subunit sequence, branching (in the case of oligosaccharides or cross-linked proteins) and substituents. Fascinating new areas of MS research include conformational analysis of biopolymers and structural studies of non-covalent complexes, which may be preformed before introduction into the MS ion source, or may be formed between trapped ions. A survey of this rapid growth will be presented, illustrated by examples from current investigations.
