catalog of human genes and disorders Information National Heart, Blood and Lung Institute cardiovascular information American Heart Association information on heart disease and stroke ATHEROSCLEROSIS is a disease that can affect people at any age, although it usually doesn't pose a threat until people reach their forties or fifties. It is characterized by a narrowing of the arteries caused by cholesterol-rich plaques of immune-system cells. Key risk factors for atherosclerosis, which can be genetic and/or environmental, include: elevated levels of cholesterol and triglyceride in the blood, high blood pressure and cigarette smoke. A protein called apolipoprotein E, which can exist in several different forms, is coded for by a gene found on chromosome 19. It is important for removing excess cholesterol from the blood, and does so by carrying cholesterol to receptors on the surface of liver cells. Defects in apolipoprotein E sometimes result in its inability to bind to the receptors, which leads to an increase a person's blood cholesterol, and consequently their risk of atherosclerosis. Currently, a debate is raging over how the various mutated forms of apolipoprotein E effect the body. As a result, many of the treatments proposed remain in their experimental phase. While mice are proving useful for modeling the human disease, a great deal of research is still required before we can fully understand the mechanisms that regulate the levels of lipoproteins - like apolipoprotein E - in the blood.

"Athero"-sclerosis is a disease in which fatty material is deposited on the wall of your arteries, which narrows the arteries and eventually restricts blood flow. It is one of several types of "Arterio"-sclerosis, a disease characterized by thickening and hardening of artery walls. However, the word "atherosclerosis" is often used to indicate any of the forms of arteriosclerosis. Fat cholesterol , and other substances accumulate in the walls of arteries and form "atheromas" or plaques. The purpose of this elegant and complex plumbing system is to deliver oxygen and nutrients to peripheral tissues and remove carbon dioxide and wastes and transport them to the excretory organs. Diffuse neurons in the lower brain initiate, inhibit, or modulate appropriate cardiovascular responses to maintain blood flow to the tissues. What is atherosclerosis?
Atherosclerosis is the buildup of fatty deposits called plaque on the inside walls of arteries. Arteries are blood vessels that carry oxygen and blood to the heart, brain, and other parts of the body. As plaque builds up in an artery, the artery gradually narrows and can become clogged. As an artery becomes more and more narrowed, less blood can flow through. The artery may also become less elastic (called "hardening of the arteries"). Atherosclerosis is the main cause of a group of diseases called cardiovascular diseases - diseases of the heart and blood vessels. Millions of Americans are counting cholesterol, spending more time on the stairmaster and less in front of TV, and trading their burgers for broiled fishall in an effort to prevent heart attacks and strokes. Doctors and scientists are doing their part, too. Their biomedical research is unraveling the complex causes of these deadly disorders, which kill almost half the people who die every year in the United States, Japan, and Europe.
Some of the most important progress has been in research on a disease called atherosclerosis. Doctors know atherosclerosis all too well as the main cause of heart attacks and strokes. In the last decade, researchers investigating what causes the disease have discovered that the endothelium, an extremely thin lining of the walls of the body's arteries, plays a crucial role. Recent discoveries about how that lining operates are providing doctors new tools for preventing atherosclerosis, diagnosing it if it occurs, and treating it effectively.

The Division of Cardiovascular Medicine has research programs in prevention and behavior, epidemiology, thrombosis, interventional technique, atherosclerosis, cardiac arrhythmias, gene therapy and nuclear cardiology. The Division has been involved in numerous NIH-sponsored research trials and studies including Thrombolysis in Myocardial Infarction (TIMI), Bypass Angioplasty Revascularization Investigation (BARI), Cardiac Arrhythmia Suppression Trial (CAST), Rapid Early Action for Coronary Treatment (REACT), Multicenter Unsustained Tachycardia Trial (MUSTT), Mode Selection for Sinus Node Dysfunction (MOST) and Worcester Area Trials for Cholesterol and Hyperlipidemia (WATCH). Current research funding includes multiple research grants from the National Institutes of Health, American Heart Association, and non-federal sources.
