Fanconi Aplastic Anemia-a lethal genetic blood/bone marrow disease- it is a non-discriminating genetic disease. The only treatment is a bone marrow transplant. Each parent must be a carrier of the gene in order to pass the gene on to his or her child. Known Italian families this illness has presented itself.

Executive Summary The Workshop on Research Directions in Fanconi Anemia was held on September 18-20, 1997 at the Columbia Inn Hotel and Conference Center in Columbia, Maryland. This important workshop resulted in attendance of individuals from across the nation and worldwide. Brief highlights from the scientific sessions and the major recommendations from the workshop are presented below. Fanconi anemia is an autosomal recessive bone marrow failure bone marrow syndrome characterized by pancytopenia, developmental defects, DNA cross linking agent hypersensitivity and cancer susceptibility. Many FA patients can be identified at birth because of congenital anomalies, although approximately 25% do not have birth defects. FA is a clinically heterogeneous disorder and can currently be divided into a least five different complementation groups designated A through E. The two cloned FA genes, FAA and FAC, encode proteins that are unrelated to each other and to other proteins in Genbank. Current research focuses on the identifying the role FA genes play in normal cells, identification and cloning new FA genes, and examining current therapies for the treatment of FA and were major topics at the workshop.

Fanconi Anemia (FA) is a recessive cancer susceptibility syndrome. Six FA genes have been cloned and encode proteins involved in a DNA-damage response pathway. In the June 13 ScienceXpress , Niall Howlett and colleagues report the characterization of mutations in cells from the FA subtypes B and D1 ( ScienceXpress 2002, DOI:10.1126/science.1073834). They discovered biallelic mutations in the breast cancer susceptibility gene; the mutations create frameshifts resulting in truncated BRCA2 protein. Howlett et al. show that restoring expression could rescue the phenotype of FA cells and restore resistance to DNA-damaging agents. The authors propose a model linking FA-associated genes to the regulation of a common DNA-damage response pathway. Links for this article

Thus, after a 3-months sabbatical in Buchwald's laboratory (H.J.) a collaborative research line was initiated aiming at extended complementation studies and gene cloning.
