UNIVERSITY OF MINNESOTA Center for Muscle and Muscle Disorders The functional organization of skeletal muscle. All skeletal muscle fibers possess a neuromuscular junction by which excitatory signals are transmitted from the nervous system via neurotransmitter, acetylcholine. The binding of acetylcholine to nicotinic receptors on the post-synaptic membrane of the muscle causes a local change in the voltage of the muscle membrane (sarcolemma). The voltage change affects the neighboring channels, allowing Na to enter and eventually K to flow out: this ion movement produces the action potential which propagates along the sarcolemma and inward via the invaginations of the transverse tubule system. This rapid voltage change next initiates the gating of Ca channels (i.e., dihydropyridine receptors) located within the transverse tubule which in turn, by some yet to be determined mechanism, cause the release of Ca from the sarcoplasmic reticulum via a set a channels known as the ryanodine receptors. Disorders of the Musculoskeletal System MUSCLE DISORDERS AND SPORTS INJURIES Over 600 muscles drape the human skeleton and give the body its characteristic form. They enable us to walk down the street, throw a ball, and stand up straight. The muscles we are most aware of are called skeletal or striated muscles. Skeletal muscles are most obvious to us visually (they are the muscles that lie just beneath the skin's surface) and they are also the only muscles that we can consciously flex and relax (voluntary muscles). There are three types of muscles: Striated Muscles. Capable of voluntary movement and connected to the bones by tough tissue called tendons. When you consciously move a part of your body, you use striated muscles. Smooth Muscles. Associated with many of the internal organs such as the stomach, intestines, uterus, and blood vessels. Researchers in the Medical School (Departments of Physiology , Biochemistry, Neurology, Anesthesiology , Physical Medicine and Rehabilitation, Opthalmology), Veterinary School (Departments of Pathobiology, Clinical and Population Sciences) and the Institute of Technology Biomedical Engineering and Mechanical Engineering ) have all expressed a strong interest in establishing a center to facilitate communication and collaboration. Individual participants have expertise in muscle anatomy, electrophysiology, biochemistry, molecular biology, genetics and development. Numerous faculty members have clinical expertise in either human or veterinary muscle diseases. Currently funded research projects include studies of normal muscle function, naturally occurring disorders of muscle, chemically-altered muscle function and transgenically-developed model systems of muscle disease. Despite this breadth of expertise and research, to date there has been no forum for communication, inadequate awareness of the expertise on campus, and lack of support for collaboration.
