Det Kongelige Videnskabsakademi i Stockholm har i dag uddelt Nobelprisen i kemi for 1997 til tre modtagere: Professor Jens C. Skou får den ene halvdel for opdagelsen af det iontransporterende enzym, Na , K -ATPase (natrium, kalium-stimuleret adenosintrifosfatase). Dette enzym opretholder balancen af natrium- og kaliumioner i den levende celle. Den anden halvdel af prisen deles mellem professor Paul D. Boyer fra USA og Dr. John E. Walker fra Storbritannien. Jens C. Skou, der er født i Lemvig i 1918, blev cand.med. fra Københavns Universitet i 1944 og dr.med. i 1954. Han kom til Aarhus Universitet i 1947 og blev udnævnt til professor i fysiologi i 1963. I 1977 blev han udnævnt til professor i biofysik. Han tog sin afsked fra universitetet i 1988, men har beholdt sit kontor på instituttet. Jens C. Skou har tidligere modtaget Leo-prisen, Novo-prisen, Konsul Carlsens Pris, Anders Retzius-guldmedalje fra Det svenske Lægeselskab, Eric K. Fernströms store nordiske pris, ligesom han er udnævnt til æresdoktor ved Københavns Universitet.

Walker, Medical Research Council Laboratory of Molecular Biology, Cambridge, United Kingdom; for their elucidation of the enzymatic mechanism underlying the synthesis of adenosine triphosphate (ATP); and with one half to Professor Jens C. Skou, Aarhus University, Denmark for the first discovery of an ion-transporting enzyme, Na+, K+-ATPase. The three laureates have performed pioneering work on enzymes that participate in the conversion of the "high-energy" compound adenosine triphosphate (ATP).
ATP synthase - an exceptional molecular machine
The German chemist Karl Lohmann discovered ATP in 1929. Adenosine triphosphate (ATP) consists of the nucleoside adenosine linked to three phosphate groups. On removal of the outermost phosphate group, adenosine diphosphate (ADP) is formed while at the same time the energy released can be employed for other reactions. Conversely, with the help of energy, an inorganic phosphate group can be bound to ADP and form ATP. Considerable quantities of ATP are formed and consumed. At rest, an adult converts daily a quantity of ATP corresponding to about one half body-weight, and during hard work the quantity can rise to almost a ton. Most of the ATP synthesis is carried out by the enzyme ATP synthase. At rest Na+, K+-ATPase uses up a third of all ATP formed.

The discovery could lead to more accurate atomic clocks for use in space navigation. Chemistry: Paul D. Boyer (U.S.), Jens C. Skou (Denmark), and John E. Walker (U.K.) for their discoveries about a molecule that allows the human body to store and transfer energy between cells. Skou received half the prize money for his discovery of an enzyme that works with adenosine triphosphate (ATP) to regulate levels of potassium and sodium in cells. Dr. Stanley Prusiner of the University of California, San Francisco, received the 1997 Nobel in physiology or medicine for his discovery of an unusual class of infectious particles called prions. Dr. Paul Boyer of the University of California, Los Angeles, is one of three recipients of the 1997 Nobel in chemistry. Boyer, a UCLA biochemistry professor emeritus, has received grant support from NIH since 1948. The vast majority of the more than $4.4 million he has received came from the National Institute of General Medical Sciences, with additional funding from the National Institute of Diabetes and Digestive and Kidney Diseases. He and Dr. John E. Walker of Cambridge, England, shared half of the Nobel in chemistry "for their elucidation of the enzymatic mechanism underlying the synthesis of adenosine triphosphate (ATP)." The other half of the chemistry prize went to Dr. Jens C. Skou of Aarhus University in Denmark for "the first discovery of an ion-transporting enzyme, sodium potassium-stimulated adenosine triphosphatase." Skou received grant money from NINDS in the early 1960's.

Long-Time NIH Grantee Wins Nobel Prize in Chemistry

Dr. Jens C. Skou of Aarhus University in Denmark, one of the other prize recipients, received support from NIH's National Institute of Neurological Disorders and Stroke in the early 1960's. "The Nobel Prize recognizes Dr. Boyer for a lifetime spent investigating the molecular mechanism by which ATPthe cellular energy that drives all biological reactionsis formed," said Dr. Marvin Cassman, NIGMS Director. "Determining how ATP is synthesized has been a central problem in biology for decades and is at the core of an understanding of metabolic processes in the cell. Dr. Boyer has been a key contributor to the molecular understanding of ATP synthesis." Since the early 1950's, Boyer has sought to understand the inner workings of ATP synthase, the enzyme he calls the "splendid molecular machine" that produces ATP. The enzyme is a complex protein rooted in the inner membranes of cellular structures called mitochondria, the "powerhouses of the cell." Over several decades, Boyer developed a model of how the various subunits of the enzyme work together like gears, levers, and ratchets to generate cellular energy. His theories were confirmed in 1994 with a detailed, three-dimensional structure of the enzyme determined by the third recipient of the chemistry prize, Dr. John E. Walker of the Medical Research Council Laboratory of Molecular Biology in Cambridge, UK. The techniques Boyer developed are now commonly used to study other ATP-utilizing enzymes.
