At the time, Fred Sanger was the Divisional head. Part of my efforts as a protein chemist was to validate his new methods for sequencing DNA by sequencing the proteins directly, and to use protein information to interpret the DNA sequences. Thus, we demonstrated the existence of triple overlapping genes in bacteriophage G4 and the modified genetic code in mitochondria. During the DNA sequencing of the genomes of human and bovine mitochondria, I became interested in the enzymes of oxidative phosphorylation. Therefore, since 1978, I have concentrated on studying the structures and functions of the ATP synthase (in bacteria, chloroplasts and mitochondria), and complex I (NADH: ubiquinone oxidoreductase) from mitochondria. Since 1986, I have also collaborated closely with the group of Ferdinando Palmieri at the University of Bari in Italy in studying a family of metabolite transport proteins in the inner membranes of mitochondria.

Walker, John E. I was born in Halifax, Yorkshire on January 7th, 1941 to Thomas Ernest Walker and Elsie Walker (née Lawton). My father was a stone mason, and a talented amateur pianist and vocalist. I was brought up with my two younger sisters, Judith and Jennifer, in a rural environment overlooking the Calder valley near Elland, and then in Rastrick. I received an academic education at Rastrick Grammar School, specializing in Physical Sciences and Mathematics in the last three years. I was a keen sportsman, and became school captain in soccer and cricket. In 1960, I went to St. Catherine's College, Oxford, and received the B.A. degree in Chemistry in 1964. In 1965, I began research on peptide antibiotics with E. P. Abraham in the Sir Willian Dunn School of Pathology, Oxford, and was awarded the D. Phil. degree in 1969. During this period, I became aware of the spectacular developments made in Cambridge in the 1950s and early 1960s in Molecular Biology through a series of programmes on BBC television given by John Kendrew, and published in 1966 under the title "The Thread of Life". These programmes made a lasting impression on me, and made me want to know more about the subject. Two books, "Molecular Biology of the Gene" by J. D. Watson, first published in 1965, and William Hayes' "Bacterial Genetics" helped to assuage my appetite for more information.

Dr John Walker MRC Dunn Human Nutrition Unit, Cambridge UK Structural and Mechanistic Studies of Mitochondria 1. ATP synthase The ATP synthase from mammalian mitochondria is a membrane bound assembly of 16 unlike proteins. It uses the energy of the electrochemical proton gradient across the inner mitochondrial membrane to make ATP. Synthesis of ATP from ADP and phosphate takes place in the extramembranous F domain by a mechanism that involves the rotation of the central a-helical structure in the g-subunit. This structure extends through the stalk linking the F domain to the membrane (known as F where the motor that generates rotation is found. We are attempting to understand the molecular mechanism of the enzyme by establishing its atomic structure by X-ray crystallography (in collaboration with Dr Andrew Leslie of the MRC Laboratory of Molecular Biology), by electron microscopy, and by mutating key amino acids in the enzyme from Saccharomyces cerevisiae. With Professor D. Mueller, Chicago, we are studying the effects of the mutations on the activity and the structure of the enzyme.
