Berg, Paul (1926-)

American biochemist who studied the techniques of cutting DNA using gene-sequence specific enzymes, originally innovated by Nathans and Hamilton Smith . Berg was able to improve the technique so that he could create new patterns of genes. This technique is known as recombinant DNA and is the method through which genetic engineering is practiced. Berg shared the 1980 Nobel Prize in chemistry with Sanger and Walter Gilbert. Nathans Sanger Smith (Hamilton)

Berg, Paul
US molecular biologist. In 1972, using gene-splicing techniques developed by others, Berg spliced and combined into a single hybrid the DNA from an animal tumour virus (SV40) and the DNA from a bacterial virus. For his work on recombinant DNA, he shared the 1980 Nobel Prize for Chemistry.
Berg was born in New York and educated at Pennsylvania State University and Case Western Reserve University. Between 1955 and 1974 he held several positions at Washington University.
In 1956 Berg identified an RNA molecule (later known as a transfer RNA) that is specific to the amino acid methionine. He then perfected a method for making bacteria accept genes from other bacteria. This genetic engineering can be extremely useful for creating strains of bacteria to manufacture specific substances, such as interferon. But there are also dangers: a new, highly virulent pathogenic microorganism might accidentally be created, for example. Berg has therefore advocated restrictions on genetic engineering research.

A dominant theme of research in his laboratory during the past ten years has been the development and application of recombinant DNA and related methodologies for the analysis of genetic recombination in eukaryotic cells. An important goal in cell and molecular genetics of mammalian cells is to be able to target exogenous genes to specific loci in the genome. This has been achieved although the frequency is low. His laboratory's current program aims to discover more about the process of gene conversion, a non-reciprocal recombination process in which sequences from one duplex DNA replace a corresponding region in a homologous duplex DNA. Both in vitro and in vivo experiments are being pursued to explore this reaction, particularly the isolation of yeast mutants blocked in their ability to carry out gene conversion between episomal and chromosomal homologous sequences. His laboratory is exploring this strategy for targeting alterations in chromosomal genes of embryonic stem cells with a goal of creating mutant animals. The HIV-1 virus induces an immunodeficiency disease in patients it infects by depleting the T-helper population. They are exploring the signal transduction pathway from the T cell receptor as a possible target for the action of one of the viral genes.

aul Berg assembled the first DNA molecules that combined genes from different organisms. Results of his experiments, published in 1972, represented crucial steps in the subsequent development of recombinant genetic engineering. By stepwise methods such as he devised, individual genes could be isolated and inserted into mammalian cells or into such rapidly growing organisms as bacteria. The genes themselves could then be studied, and their protein products expressed and even manufactured in quantity.

In creating hybrid DNA molecules, Berg employed the much-studied SV40 monkey virus and a bacterial virus known as the l (or lambda) bacteriophage. The SV40 virus has few genes, lacks a protein coat, and is is convenient to work with. The l bacteriophage normally invades a type of E. coli

    On June 19, 1975 a headline in the Rolling Stone Magazine said, "140 scientists ask, 'Now That We Can Rewrite the Genetic Code, What Are We Going to Say? Beneath that headline, the reporter, Michael Rogers, described one of the most contentious meetings in the history of science: the Asilomar Conference to debate the risks of newly discovered recombinant DNA techniques and to consider placing restrictions on them. Now although that meeting and its consequences remain controversial, its intrepid organizer, Paul Berg, our next speaker, won wide respect for his courage and articulate leadership. We are again calling upon both his bravery and his clarity to review the central tenets of molecular genetics, in a sense the intellectual fallout of what you've just heard from Jim Watson. Now this a field in which Paul has played several prominent roles as documented by many honors including the Nobel Prize in chemistry. Like many of our speakers Paul is a professor at the Stanford University School of Medicine where he directs the newly constructed Beckman Center for Molecular Medicine .     Paul Berg
    Paul Berg has made a lifelong commitment to science and to instilling his enthusiasm for research in his students. Berg, who is Cahill Professor of Cancer Research at Stanford University School of Medicine and Director of the Beckman Center for Molecular and Genetic Medicine, is renowned for his research that led to the Nobel Prize in Chemistry in 1980. Having now served as ASCB Public Policy Chair for two years, Berg noted that "being the Chair of the ASCB Public Policy Committee is a great deal easier than some, because the staff does such a good job of seeing that it runs smoothly. I am pleased to be part of the ASCB Public Policy Committee’s great traditions. They are such a great bunch of people." Berg is keenly aware of the necessity to ensure adequate funding for biomedical research and is continuously pleased with the support key Congress people have given for basic biomedical research. Despite Berg’s continued tireless work in his lab and in public policy, he does take time out to have fun. He likes to play tennis with Stanford friends—his back permitting. Pfeffer, a player herself, concedes of her elder colleague that "he plays a terrific game of tennis." Berg also enjoys collecting contemporary American sculpture and paintings. He and his wife of 50 years, Millie, have one son, John, whose profession is in music and graphic design. What is most clear from talking to Paul Berg is that he loves where he lives and what he does. He says he has lived at Stanford for over 35 years because, "it is the greatest place to live and Stanford is one of the best universities in the world." He also notes loyally that Stanford fits his personality and interest in young people because, "it is a school that promotes experimentation in education, research, and lifestyles."

    Mr. Chairman, Members of the Committee, thank you for inviting me to testify before you on this most important issue. I have followed the debate on the cloning questions we will address today and I welcome the opportunity to weigh in with my own views on the matter. It is also a distinct privilege to join Mr. Reeve, who has been such an articulate spokesman for support of research that could contribute therapies and cures for debilitating diseases, spinal cord injury being a most personal issue.
