    KISV PFT/ ... Nobel Prize in Physiology and Medicine // 1997: a new biological principle of infection // how are virus-infected cells recognized Viral infections require the virus to enter a new cell, multiply, and then to be released to infect further cells. These processes often involve changes in the structures of the viral proteins, and our goal is to determine their conformations at the various stages of an infection. Our research helps not only to explain how the virus functions, but also to design anti-viral drugs more effectively.
    The central theme of our research is to understand the structure and function of macromolecular interactions relevant to viral therapy vector design . A fascinating property of viruses is that their structural proteins have the capacity to function both in assembly and in disassembly. This paradox is made possible through a builtin flexibility among many viral proteins, in which an external or internal cellular event may trigger conformational changes. Therefore, we observe virus particles at various states of their life cycle to analyse either their assembly-intermediates or mature forms. Similarly, virus particles can be treated with numerous conditions, e.g. low pH mediated membrane fusion, to observe the key steps of viral attachment and entry into the host cell.
    Consequently, the improvement of molecular imaging, digital data processing, and advanced computing would provide better tools for observing the dynamic event and various forms of biological molecules in the virus life cycle. One of our goals is to apply the knowledge gained from the viral structures to more effective drug or vaccine design against viral infection. Structural information also faciliates the biomedical research in the design of gene therapy vectors involving virus macromolecules.

    The Ph.D. Program in Virology was formed in 1983 and is conducted under the auspices of the Division of Medical Sciences (DMS), which is part of the Graduate School of Arts and Sciences (GSAS). Ph.D. degrees are awarded through the Faculty of Arts and Sciences of Harvard University (HU). The Program currently has a total 53 students. Virology students pursue their study and research amid 600 graduate students from other programs in the Division of Medical Sciences. This program is a joint effort of 50 Virology faculty.     The name's Reed. Walter Reed: Major, US Army Medical Corps. I need your help. It's June 25th, 1900, and I've just been posted to Cuba after the end of the Spanish-American war. We lost more men to disease than bullets in that campaign. One disease in particular. In Panama, our boys have started digging a canal so that our warships can get from the Atlantic to the Pacific. Some canal! We've got big trouble - hundreds of our men have died so far from that damn disease. We call it "yellow jack" (yellow fever) . They get a high fever and jaundice. Up to half of those who show symptoms die. It's my responsibility to find out what's causing it and put a stop to it. Like I said, I need your help. The surgeon general has put three assistant surgeons under my command: James Carroll, Jesse W. Lazear and Aristides Agaramonte. We've got a few ideas so far about what could be causing the disease: What do you think? Click on one of the choices above.

    Scanned images are of West Nile virus isolated from brain tissue from a crow found in New York. The tissue was cultured in a Vero cell for a 3-day incubation period. The Vero cells were fixed in glutaraldehyde, dehyrated, placed in an Epon resin, thin sectioned, placed on a copper grid, and stained with uranyl acetate and lead citrate. The grids were then placed in the electron microscope and viewed. Total magnifications, image A: 65,625x;
