Friedreich's ataxia (FA) is a severeneurodegenerative condition with an incidence of1:50,000 in the European population. The defectiveprotein encoded by the FRDA gene is known asfrataxin. While the precise role of human frataxinremains to be determined, it appears to be involvedin regulating mitochondrial Fe. We examined frataxin in Femetabolism by implementing amodel of erythroid differentiation. Since decreasedfrataxin expression leads to mitochondrial Fe- loading in FA,our data suggest that reduced frataxin expressionresults in mitochondrial Fe sequestration for hemebiosynthesis. These findings suggest that the use ofspecific Fe chelators may have potential in thetreatment of this disease.In order to develop effective (Fe) chelators forFA, we synthesized and characterised three classesof aroylhydrazone analogues basedon: (1) 2-pyridylcarboxaldehyde isonicotinoylhydrazone (PCIH), (2) 2-quinoline carboxaldehydeisonicotinoyl hydrazone (QCIH), and (3) di-2- pyridylketone isonicotinoyl hydrazone (PKIH).
