Book Description
This book presents a state-of-the-art summary of the applications of low temperature to clinical situations, together with details of the underlying principles of biology. It provides specific information for the clinicians and research workers in a number of areas of current interest and attempts to provide a unifying theme of cryobiology of interest and value to those researching a clinical problem. Specific topics discussed include the effects of low temperatures on mammalian systems in the absence of ice and how the changes can be modulated to achieve desired results; low temperature storage of tissues and organs for transplantation in the liquid state; the effects of whole body hypothermia in man and how it relates to mammal hibernation; problems associated with ice formation and the subsequent freezing of cells and tissues; cryopreservation of blood cells, reproductive cells, and tissues, such as the skin and cornea. Other interesting issues featured include the developments in cryopreservation of large, highly-organized structures and the destructive powers of ice formation in cryosurgery of diseased tissues. ... Description:
Fish embryos have never been successfully cryopreserved because of the low permeability of cryoprotectants into the yolk. Recently, we used aquaporin-3 fused with a green fluorescent protein (AQP3GFP) to modify the zebrafish embryo, and demonstrated that the pores functioned physiologically. This increased the water and cryoprotectant permeability of the membranes. We have continued our work on AQP3-modified embryos and here we report their developmental expression of AQP3, the success of various culture media on their survival and development, and their reproductive success. The AQP3GFP expression begins within 30 m after the mRNA AQP3GFP injection into the yolk of the 1- to 4-cell embryo. This expression is distributed in the membranes throughout the blastoderm and the yolk syncytial layer within 24 h. It diminishes after 96 h. We found no difference in the survival or normal development of embryos from AQP3GFP or wild-type adults. Additionally, zebrafish embryos did not require special culture medium to survive after AQP3GFP modification. In fact, they survived best in embryo medium (ca. 40 mOsm). Embryos reared entirely in embryo medium had a higher percent survival and a higher percent normal development than those exposed to a high osmolality sucrose culture medium (ca. 330 mOsm). The mechanism whereby these embryos can maintain their internal osmolality in a hypoosmotic solution with water channels in their membranes is unknown. ... Read more

Book Description
While it is barely 50 years since the first reliable reports of the recovery of living cells frozen to cryogenic temperatures, there has been tremendous growth in the use of cryobiology in medicine, agriculture, horticulture, forestry, and the conservation of endangered or economically important species. As the first major text on cryobiology in the genomic era, Life in the Frozen State describes the current understanding of how living cells and complex organisms survive very low temperatures. Leading world experts combine fundamental theory and practice across a spectrum of species and applications to evaluate how cryobiology can benefit humanity. Chapters encompass disciplines ranging from mathematical modeling and biophysics, to the molecular biology of stress gene expression and the clinical banking of cells and tissues. This book provides a unique opportunity to explore the subject in a multidisciplinary context, which has historically been the key to realizing some of the most exciting advances in low temperature research.Features· Integrates fundamental theory and practice across a broad range of species and applications · Discusses cryobiology within a multidisciplinary context · Emphasizes how the current knowledge of cryobiology can be applied to benefit humanity through health care and conservation ... Book Description
Moving rapidly from science fiction to science fact, cryopreservation is an integral part of many research, development, and production processes in industry and academia. The preservation sciences have emerged as an interdisciplinary platform that incorporates the fundamentals of cell and molecular biology, and bioengineering, with the classic methodological approaches to freezing and drying living matter. Researchers and biotech companies must scramble to learn about, and adopt, the new preservation strategies that incorporate the classic and the new molecular-based approaches. The first reference of its kind, Advances in Biopreservation addresses the breadth of the field of biopreservation. Written by recognized international leaders in biopreservation, the book expertly integrates recent molecular-based discoveries with classically-based biopreservation basics, offering a new look at advances in the emerging regenerative medicine arena. In addition to general concepts of biopreservation, it includes chapters detailing subspecialty principles, such as biologically active macromolecules, stem cells, and gene therapy, as well as new protocols for cell, tissue, and organ preservation. The classically based approaches to biopreservation are now recognized as bottlenecks to advances in the emerging regenerative medicine arena. Recent discoveries have changed the basic foundations upon which the preservation sciences were built. Integrating the concepts of first generation cryopreservation with the new paradigm, this book is a fundamental reference to the latest advances in the field. ... Read more
