Product Description
Webster's bibliographic and event-based timelines are comprehensive in scope, covering virtually all topics, geographic locations and people. They do so from a linguistic point of view, and in the case of this book, the focus is on "Cryobiology," including when used in literature (e.g. all authors that might have Cryobiology in their name). As such, this book represents the largest compilation of timeline events associated with Cryobiology when it is used in proper noun form. Webster's timelines cover bibliographic citations, patented inventions, as well as non-conventional and alternative meanings which capture ambiguities in usage. These furthermore cover all parts of speech (possessive, institutional usage, geographic usage) and contexts, including pop culture, the arts, social sciences (linguistics, history, geography, economics, sociology, political science), business, computer science, literature, law, medicine, psychology, mathematics, chemistry, physics, biology and other physical sciences. This "data dump" results in a comprehensive set of entries for a bibliographic and/or event-based timeline on the proper name Cryobiology, since editorial decisions to include or exclude events is purely a linguistic process. The resulting entries are used under license or with permission, used under "fair use" conditions, used in agreement with the original authors, or are in the public domain. ... Read more

Description:
Fish embryos have never been successfully cryopreserved because of the low permeability of cryoprotectants into the yolk. Recently, we used aquaporin-3 fused with a green fluorescent protein (AQP3GFP) to modify the zebrafish embryo, and demonstrated that the pores functioned physiologically. This increased the water and cryoprotectant permeability of the membranes. We have continued our work on AQP3-modified embryos and here we report their developmental expression of AQP3, the success of various culture media on their survival and development, and their reproductive success. The AQP3GFP expression begins within 30 m after the mRNA AQP3GFP injection into the yolk of the 1- to 4-cell embryo. This expression is distributed in the membranes throughout the blastoderm and the yolk syncytial layer within 24 h. It diminishes after 96 h. We found no difference in the survival or normal development of embryos from AQP3GFP or wild-type adults. Additionally, zebrafish embryos did not require special culture medium to survive after AQP3GFP modification. In fact, they survived best in embryo medium (ca. 40 mOsm). Embryos reared entirely in embryo medium had a higher percent survival and a higher percent normal development than those exposed to a high osmolality sucrose culture medium (ca. 330 mOsm). The mechanism whereby these embryos can maintain their internal osmolality in a hypoosmotic solution with water channels in their membranes is unknown. ... Read more

Product Description
While new discoveries have led to much dramatic growth in cryo-electron microscopy, researchers will never be able to take full advantage if they lack access to the details that make these techniques understandable and applicable.

The Handbook of Cryo-Preparation Methods for Electron Microscopy provides researchers with a complete reference that will show them how to equip their labs with the right materials and methods to take full advantage of the latest advances. This pioneering work brings together a group of internationally renowned researchers, some the very inventors of the methods they describe, to share their knowledge and recipes. Taking care to explain the history behind the techniques and to demonstrate their use, this book presents the latest theory, principles, and protocols supplemented by hundreds of illustrations.

Contributions to this handbook –

Ö         Describe cryo-methods aimed at perfect preservation for fine structural analysis

Ö         Teach how to arrest physiological processes by cryo-fixation

Ö         Reveal the secrets for high-resolution snapshots of life by CEMOVIS

Ö         Cover applications of cryo-methods such as electron crystallography, 2D/3D structure analysis, protein localization, and cryo-electron tomography

Ö         Demonstrate the use of vitreous water as an intermediate step for localization of biological ions and molecules

Ö         Present hybrid methods of freeze-substitution and freeze-drying for immunolabeling and determining molecular geography

Ö         Illustrate freeze-fracture, cryo-ultramicrotomy, and resin embedding

Ö         Highlight the Tokuyasu method and the new rehydration technique for highly efficient immunolabeling

Ö         Include summary tables to compare and obtain appropriate criteria

Includes a wealth of immediate reference material for daily use

With clear, step-by-step recipes and lists of tools, ingredients, and suppliers, this handbookprovides researchers with the knowledge and techniques to adopt the latest cryo-methods to their current research. ... Read more

Editorial Review

Product Description
Cryobiology is the study of living things at low temperatures. Refrigeration or freezing prevents the decay of living things, but science has recently begun to explore whether cold not only prevents decay, but also preserves life. An interdisciplinary subject that combines fundamental research on organisms adapted to natural freezing stresses and emerging areas of applied biotechnology and biology, cryobiology is starting to play an increasingly important role in medicine, agriculture, horticulture and forestry, as well as the conservation of endangered or economically important plant, animal and microbial species. Life in the Frozen State is a comprehensive review of the effects of freezing on living organisms. It evaluates how this knowledge can be applied to benefit humanity through advances in healthcare and conservation. A major feature of the book is the combining of fundamental theory and practice across a wide range of species and applications. This provides a unique opportunity to explore the subject in a multidisciplinary context. Historically, some of the most exciting advances in low temperature research have come from cross-discipline interaction, and this book provides a timely stimulus for further advances in cryobiology. ...
