Product Description
Direct methods are now used in a variety of cases: X-ray, neutron or electron data; single crystal and powder data; small molecules and macromolecules. While direct methods solved in practice the phase problem for small molecules, their application to macromolecules is recent and still undergoing development. This book describes the fundmental methods; in particular it is shown how the methods can be optimized for powder, neutron or electron data, and how they can be integrated with isomorphous replacement, molecular replacement and anomalous dispersion techniques. Maximum entropy methods are also described and discussed. Sets of test structures are used throughout the various chapters to verify the mathematical techniques and to provide practical examples. This book offers both a comprehensive description of direct methods in crystallography and an invaluable reference tool. The first three chapters provide an introduction to the field, with sufficient material to constitute a university course. Subsequent chapters are aimed at graduate students and working crystallographers. Basic results are described and discussed in the main body of the text, while the appendices complement these with in depth mathematical details. The quoted literature is extremely wide and the interested reader can find suggestions for future work and further reading throughout the book. ... Read more

4-0 out of 5 stars A must for direct method fans.
The book has a logical structure, goes into details and is clearly written. It provides a good starting point for every person interested in the theory behind direct- and related structure solving methods. Althoughthe layout (most notably figures) and the index are of lower quality, thebook on itself is a must. ... Read more

Editorial Review

Product Description
The re-emergent field of quantitative electron crystallographyis described by some of its most eminent practitioners. They describethe theoretical framework for electron scattering, specimenpreparation, experimental techniques for optimum data collection, themethodology of structure analysis and refinement, and a range ofapplications to inorganic materials (including minerals), linearpolymers, small organic molecules (including those used in nonlinearoptical devices), incommensurately modulated structures (includingsuperconductors), alloys, and integral membrane proteins. Theconnection between electron crystallography and X-ray crystallographyis clearly defined, especially in the utilisation of the latestmethods for direct determination of crystallographic phases, as wellas the unique role of image analysis of high-resolution electronmicrographs for phase determination. Even the aspect of multiple beamdynamic diffraction (once dreaded because it was thought to precludeab initio analysis) is considered as a beneficial aid forsymmetry determination as well as the elucidation of crystallographicphases, and as a criterion for monitoring the progress of structurerefinement. Whereas other texts have hitherto preferentially dealtwith the analysis of electron diffraction and image data from thinorganic materials, this work discusses - with considerableoptimism - the prospects of looking at `harder' materials, composed of heavier atoms.
Audience: Could be used with profit as a graduate-level courseon electron crystallography. Researchers in the area will find astatement of current progress in the field. ... Read more

Product Description
The main theme of the book will be on the interactions of macromolecules and the consequences for biological function; lessons learned from the detailed study of the individual components will also be included. To bring together contributions of leading structural scientists who are addressing even more challenging problems, such as crystallization of membrane proteins, the structures of large assemblies of proteins, proteins and RNA, and proteins and DNA, and the structures of large viruses is also one of the aims of the book. In addition, advances in macromolecular crystallography have given us new eyes with which to look at biology and by working at the EM/Xray interface and by exploiting the potential of third generation synchrotron radiation sources with their extraordinarily bright beams for data collection from very small crystals new very important targets can be addressed.Te book should also give the flavour of this new frontiers. ... Editorial Review

Product Description
Due to its interdisciplinary nature, crystallography is of major importance to a wide range of scientific disciplines including physics, chemistry, molecular biology, materials science and mineralogy. However, information is currently divided amongst traditional physics, chemistry and materials science books. This book collates previously disparate literature into one comprehensive and practical source, providing a thorough understanding of the information contained in crystallographic data files and the application of x-ray diffraction methods. The book has been written for final year and postgraduate students. ... Read more
