Nicolas Gombert stammt vermutlich aus dem Dorf La Gorgue, das westlich von Lille liegt. Womöglich war Gombert, wie Heinrich Finck 1556 berichtet, ein Schüler Josquin Desprez' († 1521), der seit 1504 im nahegelegenen Condé-sur-l'Escaut Propst war. Bis zu seinem Eintritt in den Dienst Kaisers Karls V. gibt es keine gesicherten Informationen über Gombert. Doch aus dem Umstand, dass er in eine der besten Kapellen Europas aufgenommen wurde, darf auf eine hervorragende Ausbildung geschlossen werden. Am 2. Oktober 1526 ernannte Kaiser Karl V. in Granada den Kleriker Gombert zum Präbendar in Kortrijk und Béthune. Gombert diente dem Kaiser als Sänger. Bis 1529 hielt sich Gombert hauptsächlich in Spanien auf, da die Kapelle den Kaiser auf dessen Reisen begleitete. Längere Aufenthalte wurde in Toledo, Sevilla, Granada, Valladolid, Valencia, Burgos und Madrid eingelegt. 1529 ernannte Karl V. Gombert zum 'Maistre des enfants de la chapelle'.
