THEN THE DOCTOR GAVE ME A LETTER SAYING THAT INDEED HE HAD BEEN GIVEN BLOOD, BY A DOCTOR WHEN HE WAS JUST A WEEK OLD. MY HUSBAND AND I WERE SHOCKED, TO SAY THE LEAST, AND OUTRAGED ALSO, SINCE WE HADN'T EVEN BEEN TOLD, AND TO MAKE THE SITUATION EVEN MORE FRUSTRATING, OUR FAMILY DOCTOR DIDN'T HAVE A FACT SHEET, AND ALL HE TOLD US IS THAT IT IS AN INCURABLE, FATAL DISEASE.     Research interests in our laboratory include developmental neurobiology, particularly cell lineages and the regulation of developmental fates; the nature of progenitor cells in the adult CNS; and glial cell pathology in neurodegenerative disorders. We are tracing cell lineages in the developing mammalian CNS to study the origins of astrocytes and oligodendrocytes from immature neuroectodermal cells. Using retroviral mediated gene transfer, we are following the migration and differentiation of immature cells of germinal zones in vivo and have determined that there are time- and location-specific patterns of glial development. We are examining what roles environmental vs. lineage-controlled factors play in specifying cell fate. We have also been investigating the nature of cycling cells in the adult mammalian CNS, characterizing their phenotypes and their fates under normal and pathological circumstances. For example, cycling, immature cells that normally do not differentiate are stimulated to develop into myelinating oligodendrocytes after demyelination, thus contributing to the repair of the lesion. We are studying glial reactions in pathological states, focusing upon specific proteins of astrocytes. Particular interests include GFAP, the major intermediate filament type in astrocytes, and the small heat shock protein, alpha B-crystallin, expressed in glial cells normally, but upregulated in pathological conditions. Massive amounts of crystallin accumulate in the brains of children with Alexander's disease, a fatal CNS degenerative disorder, recently determined to be associated with point mutations in GFAP. We believe that alterations in cytoskeletal organization evoke a "stress" response in astrocytes and are now studying the mechanisms by which this may occur.
