    Life-Saving Cryobiology London, England, 1949. Audrey Smith and Charles Polge put a few drops of glycerin into a petri dish containing chicken sperm. The glycerin, which was intended to slow down the sperm so the scientists could study its movement, had an unexpected side effect—when the scientists accidentally froze these sperm in liquid nitrogen, they survived. This "chance observation," as the pair described it in their article in the scientific journal Nature, was a milestone—the first time in history anyone had successfully cryopreserved living mammalian cells or tissue. How did Gao, who's original dream was to design jet engines, become a cryobiologist? He answers very simply: "It's the challenge." "In your very first undergraduate biology class you can question the professor, and the professor will often tell you, "I don't know" or "It's not clear now.' But in undergraduate physics and mathematics class, you can always get an answer," Gao says. "In biology there is an enormous amount we know nothing about. It's very challenging. There are a lot of things to be discovered, and that really motivates me." A native of China, Gao received his bachelor's degree in mechanical engineering at the University of Science and Technology of China then moved to Canada to study bioengineering at Concordia University in Montreal. He earned his Ph.D. in 1991 and was a postdoctoral fellow at Methodist Hospital of Indiana in Indianapolis. Gao ultimately became director of the cryobiology and bioengineering division at Methodist Hospital. In 1998 he left that position to come to UK.

    The following article is from the Prometheus Project a good source for information on current cryonics research. I agree with them that the best and only hope for safe freezing of tissue will involve vitrification (amorphous crystal freezing) of live tissue. Without more advanced cryopreservation techniques (below), I see little hope for the ravaged corpses frozen at Alcor, Inc. But the freezing and thawing of living organisms is vital to future space travelers and those who would wish, as I do, to sleep through this dangerous and primitive era and thaw out in a century or two. Vitrification holds promise of delivering on this concept. My question is whether or not tuned ultrasound frequencies could enhance the vitrification process, shattering the ice crystal lattices without significant heating to the system.

    the study of the physiology of low temperature.
    Prior to the advent of modern medical science, it had been observed that survival from severe hypothermia was possible, if uncommon. Many animals enter states of dormancy to conserve water and energy during particularly adverse climatic conditions e.g., dormice and bears, lungfish and some desert toads on Terra. However, there is a limit to how long dormancy can be maintained. Metabolism continues, albeit at a greatly slowed rate. Long term tissue preservation requires metabolic arrest. To effect this, cell water must be locked in place to prevent cellular proteins from working. Water unfortunately expands on freezing (crystallisation), which causes cell rupture and death. It had been observed that rapid freezing limited the size of ice crystals formed. Immersion in liquid nitrogen (-196°C) was used to preserve sperm, ova, embryos, seeds and pollen as early as Tech Level 6. By Tech Level 8, it had been shown that animals such as frogs and mice could be snap-frozen, thawed and revived. Survival rates were predictably low. The great breakthroughs which usher in the modern era of low berthing take place at Tech Level 9. Hibernation 'Fast drug' is a combination of cell signalling factors and receptor blockers which are typically discovered in the course of early research into metabolism and aging. Its effect is to slow down metabolic rate a staggering sixty times. Unconsciousness ensues within two minutes of adminstration. Body temperature attains ambient levels within four to six hours. As a result, muscle rigidity is typical (below core temp 25°C). It is difficult to distinguish between fast drug administration, severe hypothermia or recent death. TL 9+ blood testing will identify fast drug residues.
