Laboratory and Facilities
T he Samuel Roberts Noble Microscopy Laboratory (SRNML) was created by a generous donation of the Noble Foundation of Ardmore, Oklahoma in 1979. Today, the Lab occupies 15 rooms for instruments, photography and instruction in the Noble Microscopy Laboratory building and six research rooms in the adjacent George Lynn Cross Hall, which is occupied by the Department of Botany and Microbiology at 770 Van Vleet Oval. The facility occupies a total of nearly 4,500 square feet for research and instruction. The SRNML is available for use by researchers at all levels (undergraduate, graduate, post-doctoral and faculty) with cooperative research opportunities with clients both on- and off-campus. The Laboratory also offers open houses and tours for elementary and high school groups and other groups, including explanations of how the instruments work, operational displays and examples of how they are used in industry. Currently, the SRNML houses the most sophisticated collection of electron microscopes in the State, with a current capital value of nearly $1.4 million. University researchers requiring the instruments may use the equipment for a nominal charge, plus reimbursement for photographic supply usage and expendables. Usage may be arranged with the Director or the Electron Microscopists.

WHAT'S NEW? SEM examination of microorganisms captured on bacterial filters produced some structures which were difficult to explain. Based on the suggestions of two microbiologists, where were assumed to be "actinomycetes". However, some fresh filters contained such structures on their surfaces. Please see new site. A suspicion about the biological nature of the filter contamination was first noticed in a study of microorganisms in garden compost. The information has now been corrected and links to other compost sites have been added. The Guests 4 section has for some time been enriched by a contribution from Dr. T. Beveridge et al. on the structure of sea buckthorn fruits. Does cryofixation of hydrated foods produce more accurate images of their structures than chemical fixation? It may, under certain conditions, which, however, are not easy to meet. Otherwise the structure of the sample may be distorted by the development of ice crystals.

What are Electron Microscopes?
Electron Microscopes are scientific instruments that use a beam of highly energetic electrons to examine objects on a very fine scale. This examination can yield the following information:
Topography
The surface features of an object or "how it looks", its texture; direct relation between these features and materials properties (hardness, reflectivity...etc.)
Morphology
The shape and size of the particles making up the object; direct relation between these structures and materials properties (ductility, strength, reactivity...etc.)
Composition
The elements and compounds that the object is composed of and the relative amounts of them; direct relationship between composition and materials properties (melting point, reactivity, hardness...etc.)
Crystallographic Information
How the atoms are arranged in the object; direct relation between these arrangements and materials properties (conductivity, electrical properties, strength...etc.)

Aperture
Bright Field Imaging
A imaging mode in a TEM that uses only Unscattered Electrons to form the image. Contrast in such an image is due entirely to thickness and density variations in a sample.
Backscattered Electron
Produced by an incident electron colliding with an atom in the specimen which is nearly normal to the incident's path. The incident electron is then scattered "backward" 180 degrees with no appreciable loss of energy, an elastic collision.
Backscattered Electron Imaging
The production of backscattered electrons varies directly with the specimen's atomic number. This differing production rates causes higher atomic number elements to appear brighter than lower atomic number elements. This interaction is utilized to differentiate parts of the specimen that have different average atomic number. (See Interaction Volume) An example is shown in the SEM output section, specifically the mechanically alloyed specimen micrograph. Detection of Backscattered Electrons is achieved by using a "donut" shaped solid state semiconductor device mounted on the bottom of the objective lens. When Backscattered Electrons strike the detector electron-hole pairs are created which are then counted. This quantity is translated into a pixel intensity and displayed on the CRT, forming the image. See the

Welcome to my website
Finally I started to set up my image gallery where I am presenting pictures of various lymphotropic herpesviruses using electron microscopy, immuno electron microscopy and immuno fluorescence microscopy. I am going to add more pictures every week or when I get time for scanning in the pictures. These pictures were taken on cell lines available in my laboratory and using monoclonal antibodies developed in my lab. The herpesvirus information page will be added soon. The Flash 4 program ( virtual microscope) for fluorescence and electronmicroscopic pictures will be added (finally it start to work), which will allow to view the pictures, presented in the image gallery, at various magnifications and to move around in the picture.
