Extractions: For many biologists, the microscope belongs to the gadgets they use most. The reason is obvious: cells are the basic units of life and their size is, with only few exceptions, below the upper limit of the resolution of the human eye. Numerous statements on the refraction of light by lenses or waterfilled spheres and the resulting magnification proof, that the effects of magnification were already known in antiquity. In the 13th century, the British natural scientist ROGER BACON used segments of glass spheres as magnifying glasses and recommended them to partially sighted persons as spectacles. The grinding of lenses was perfected in the following centuries. The physician P. BORCH records, that HANS and ZACHARIAS JENSEN (father and son) from the Dutch town Middelburg invented the compound microscope at the beginning of the 17th century by placing one lens behind the other. They are regarded as the inventors of the telescope, too. Other sources name C. J. DREBBEL (1572 - 1634) from Alkmaar as the inventor of the microscope. Even if this should not be the case, he has helped a lot to make the microscope known. The term microscope was introduced by members of the Italian Accademia de Lincei (Academy of The Lynx), whose most prominent member was G. GALILEI.

In the post genomic era, fluorescence imaging provides critical information on the functions of proteins and on the dynamics of multi-protein structures. Our ability to image cellular structures has been revolutionized by methods for tagging proteins with naturally fluorescent proteins such as GFP and by the development of advanced digital microscopes. Image informatics However. no universally accepted formats exist for storing and sharing multi-dimensional image data (having spatial, temporal and spectral range) and for preserving the all-critical metadata that records what the experiment is about. Even more importantly, relatively few analytical routines are available for extracting quantitative data from images and linking them to other biological information. Developing these tools is the goal of image informatics. OME The Open Microscopy Project (OME) is an open source software project to develop a database-driven system for the quantitative analysis of biological images. Extractions: The Center for Electron Microscopy (CEM) at Southern Illinois University is a service unit supported by the Office of Research Development and Administration (ORDA). The CEM provides service and training in electron and light microscopy. This is accomplished by means of graduate level courses in scanning (SEM) and transmission electron microscopy (TEM) as well as training in the use of all instrumentation in the CEM. Trained staff will prepare specimens and assist in the viewing of samples and assist in the writing of technical or budgetary proposals related to electron microscopy and the writing of reports for granting agencies. CEM sponsors workshops on modern research methods in electron microscopy and offers tours of the facility to the university and community. The internal rates are designed to recover all costs associated with expendible supplies, service contracts on major instrumentation and a portion of the salaries of technical personnel. The CEM is available for outside grant and contract work. Outside rates are negotiated through the Office of Research Development and Administration and are designed to recover all operating expenses as well as an established overhead rate. Major equipment consists of a state-of-the-art analytical system composed of a Hitachi 7100 TEM equipped for x-ray analysis (NORAN Voyager III) and digital, high resolution imaging at 125 kV. The TEM is connected to a variable pressure SEM also with x-ray analysis capability. This permits researchers to observe non-conductive specimens under more natural conditions than the conventional SEM and TEM (also available). The EM system is networked with various light microscopes and macroscopes capable of capturing low light level images for further analysis. Preparatory equipment includes: ultramicrotomes, vacuum evaporators, critical point and freeze dryers, sputter coater, specimen trimmers, darkroom set-ups, copy stands, and miscellaneous small lab equipment.
